The soybean (Glycine max) was modified for resistance to lepidopteran insects and tolerance to glufosinate herbicides. For resistance to lepidopteran pests, the soybean expresses the Bacillus thuringiensis Cry1Ac and Cry2Ab insecticidal crystal proteins, which form non-specific ion-conducting pores in the apical membrane of midgut epithelial cells, leading to disruption of the midgut epithelium and eventual death of susceptible insects. For tolerance to glufosinate, the soybean expresses phosphinothricin N-acetyltransferase (PAT), which inactivates the herbicide through acetylation.

Information on the inserted DNA sequences
The transforming plasmid pDBN4031 contained a transfer DNA (T-DNA) region comprising three gene cassettes expressing the insecticidal proteins Cry2Ab2 and Cry1Ac derived from Bacillus thuringiensis and the herbicide tolerance protein phosphinothricin N-acetyltransferase (PAT). Transformation of soybean was achieved through Agrobacterium tumefaciens-mediated transformation.
(1) Transcription of the cry2Ab2 coding sequence is directed by the Actin 2 promoter from Arabidopsis thaliana. The coding sequence is preceded by the CTP2 chloroplast transit peptide sequence from Arabidopsis thaliana, which directs the expressed protein to the chloroplast. Transcription is terminated by the rbcS E9 terminator from Pisum sativum. Expression of this cassette results in production of the Cry2Ab2 insecticidal crystal protein, which confers resistance to susceptible lepidopteran insect pests.
(2) Transcription of the cry1Ac coding sequence is directed by the rbcS promoter from Arabidopsis thaliana. The coding sequence is preceded by the rbcS chloroplast transit peptide sequence from Arabidopsis thaliana, which directs the expressed protein to the chloroplast. Transcription is terminated by the nopaline synthase terminator from Agrobacterium tumefaciens. Expression of this cassette results in production of the Cry1Ac insecticidal crystal protein, which confers resistance to susceptible lepidopteran insect pests.
(3) Transcription of the pat coding sequence is directed by the Cauliflower mosaic virus 35S promoter. Transcription is terminated by the Cauliflower mosaic virus 35S terminator. Expression of this cassette results in production of the phosphinothricin N-acetyltransferase (PAT) protein, which confers tolerance to glufosinate herbicides by inactivating the herbicide through acetylation.

Molecular characterization demonstrated that the genome of DBN8205 soybean contains a single, intact copy of the cry1Ac, cry2Ab2 and pat expression cassettes.
